This kit contains KnockOut™ SR (KSR) and mouse embryonic fibroblasts (MEFs) isolated from mouse (Strain ICR) embryos at Day E13.5. KnockOut™ SR is a defined, serum-free formulation optimized to expand and maintain undifferentiated ES and iPS cells in culture. KSR directly replaces FBS in existing protocols and can be used for culturing mouse and human ESC and iPSCs on MEFs. The performance of KSR is enhanced when used with KnockOut™ DMEM, catalog #10829-018. The MEFs were expanded and inactivated by gamma irradiation and cryopreserved at passage 2 (P2). Before cryopreservation, the cells were tested for viability (>80% post-thaw viability), verified for growth arrest by plating inactivated cells at low density and monitoring for foci of cell growth after 14 days in culture. The MEFs were also assessed for their ability to support growth of mouse embryonic stem cells (mESCs) and human embryonic stem cells (hESCs) marker expression (Oct4, SSEA-1, Nanog). For optimal culture performance, we recommend culturing the MEFs in DMEM (SKU# 10569-010), with 10% Fetal bovine serum (SKU# 16000-044). Following 24 hr culture, the MEFs can be used for plating ESCs or iPSCs in KSR.
